Thanks!|||You don't have to--but if you can suck out as much of the red slime algae with your siphon as possible before you treat the tank, the treatment works better. You can also skip the water change and just remove as much of it with a net as possible, or a scrubber.
One airstone should be enough if it is a large airstone that produces lots of fine bubbles. Most airstones are crappy and produce really big bubbles or don't produce enough bubbles for adequate surface agitation and water circulation. I have always used two stones, but one should work if its a good quality airstone and your pump works good.
